چکیده

1. It should be pointed out that substances present in the test mixture act as electron transfer agents, or catalysts of the reaction which are recycled, i.e, reduced by DTT or superoxide and reoxidized by oxygen species in a cyclic manner (see Li et al 2009a reference). We do mention that the reaction is catalytic in redox-active species, on pg 11326 starting on line 10: “Oxidation of DTT by metals is a catalytic process, as it is for quinones (Kumagai et al., 2002). For example, in our experiments 1.0 μM of Cu(II) oxidizes 13.2 μM of DTT in 15 min. Cu(II) likely accepts an electron from DTT, forming Cu(I), which rapidly donates an electron to dissolved oxygen to form superoxide and re-form Cu(II).”

On dithiothreitol (DTT) as a measure of oxidative potential for ambient particles: evidence for the importance of soluble transition metals.

The rate of consumption of dithiothreitol (DTT) is increasingly used to measure the oxidative potential of particulate matter (PM), which has been linked to the adverse health effects of PM. While several quinones are known to be very reactive in the DTT assay, it is unclear what other chemical species might contribute to the loss of DTT in PM extracts. To address this question, we quantify the...

This is a very well conducted study, which shows novel findings about the role of metals in oxidative properties of ambient aerosols. The manuscript provides evidence that the DTT assay used as a measure of PM oxidative potential is not a “one species” (e.g. quinone) driven method, which it was used to be considered in research community, but rather involves multiple species. However, based on ...

Oxidant production from source-oriented particulate matter – Part 1: Oxidative potential using the dithiothreitol (DTT) assay

Recent epidemiological evidence supports the hypothesis that health effects from inhalation of ambient particulate matter (PM) are governed by more than just the mass of PM inhaled. Both specific chemical components and sources have been identified as important contributors to mortality and hospital admissions, even when these end points are unrelated to PM mass. Sources may cause adverse healt...

Oxidative potential of ambient water-soluble PM2.5 in the southeastern United States: contrasts in sources and health associations between ascorbic acid (AA) and dithiothreitol (DTT) assays

The ability of certain components of particulate matter to induce oxidative stress through the generation of reactive oxygen species (ROS) in vivo may be one mechanism accounting for observed linkages between ambient aerosols and adverse health outcomes. A variety of assays have been used to measure this so-called aerosol oxidative potential. We developed a semi-automated system to quantify oxi...

A semi-automated system for quantifying the oxidative potential of ambient particles in aqueous extracts using the dithiothreitol (DTT) assay: results from the Southeastern Center for Air Pollution and Epidemiology (SCAPE)

A variety of methods are used to measure the capability of particulate matter (PM) to catalytically generate reactive oxygen species (ROS) in vivo, also defined as the aerosol oxidative potential. A widely used measure of aerosol oxidative potential is the dithiothreitol (DTT) assay, which monitors the depletion of DTT (a surrogate for cellular antioxidants) as catalyzed by the redox-active spe...
